糖尿病是现今生活的一种常见慢性病,长期高血糖容易造成细小血管的损伤和免疫能力低下,进一步导致糖尿病人容易遭受细菌感染并久治不愈。传统抗生素的过度使用滥用引起新型耐药细菌的出现和细菌感染加重。银作为古老而传统的广谱抗菌材料且不易产生细菌耐药已受到广泛关注。与普通银相比,近年来兴起的银纳米颗粒显著增强的抗菌能力和明显降低的生物毒性已在生物材料、生物医学等领域广泛研究和使用。但常规方法得到的Ag NPs极易团聚不仅降低了其临床应用效果,长期高浓度使用也容易引起的细胞毒性。

生物学院刘斌副教授课题组针对纳米银材料的团聚和抗菌效能问题,创造性的将银纳米粒子镶嵌在具有良好光热性能的普鲁士蓝纳米复合物表面,构建了一种双模式纳米抗菌复合物,结合近红外光激光照射技术,有效地促进了糖尿病小鼠多药耐药菌感染伤口的快速愈合。体外研究显示:这种联合抗菌策略通过破坏细胞完整性、产生ROS、降低ATP、氧化GSH等途径发挥抗多药耐药菌MRSA作用;体内实验表明:该策略通过减轻局部炎症反应和诱导创面血管内皮生长因子表达来促进MRSA感染的糖尿病创面愈合。研究结果以论文形式发表在生物材料Top期刊Biomaterials(10.1016/j.biomaterials.2020.119936),并已同步申请专利(CN201911347212.6),为后续产品研发奠定了坚实的基础。论文第一单位为湖南大学生物学院,第一作者为湖南大学童春义副教授、湖南师范大学钟祥华硕士生和杨粤军教授,湖南大学刘斌副教授和湖南中医药大学王炜教授为并列通讯作者。

近年来,该研究团队在刘斌博士的带领下,围绕新型纳米生物材料在抗肿瘤、抗菌领域和天然药物靶向筛选领域中的应用开展了较系统的研究,得到了一系列具有重要理论价值和应用前景的研究工作,在BiomaterialsAnalytical ChemistryChemical CommunicationsNanomedicineBiomaterials ScienceScience Bulletin等权威期刊发表论文30多篇。
